What is publicly known is that the CIA has a department called SAC (Special Activities Center) formerly the SAD. Apparently they hire the best from Delta, DEVGRU/Seal Team Six, and other “tier 1” US military units. They can also hire from police units and even civilians in special cases. You don’t apply, they will contact you.

“Assassins” often need more than just combat skills, the CIA would be looking for people with other unique skill sets such as language, cryptography, cyber-security etc. Often the real assassins are locally developed assets not from the CIA at all, the CIA finds motivated people who has access to the target and blackmails, bribe them with visas/green cards and asylum in the US or pays them to assassinate the target. The CIA is also known to work with insurgencies and revolutionary forces to conduct assassinations.

The idea of the super spy one man assassin is likely more Hollywood than fact. Case in point they sent Seal Team Six after Bin ladin, not some super spy.
